CD172a (SIRPα) is a type I transmembrane protein with one V-set Ig-like and two C-set Ig-like domains extracellularly, and two ITIM motifs plus a proline-rich region in the cytoplasmic tail. It is expressed on monocytes, macrophages, myeloid cells, and neuronal tissue. Phosphorylation of its ITIMs recruits and activates tyrosine phosphatases PTPN6 and PTPN11, mediating negative regulation of signaling pathways. Known ligands include CD47, SP-A, and SP-D, enabling SIRPα to modulate phagocytosis, immune cell activation, and other regulatory processes.
